Simultaneous qualitative and quantitative determination of phenylethanoid glycosides and flavanoid compounds in Callicarpa kwangtungensis Chun by HPLC-ESI-IT-TOF-MS/MS coupled with HPLC-DAD†
Abstract
An effective and comprehensive method was developed for the simultaneous analysis of phenylethanoid glycosides and flavanoid compounds in the ethanol extracts of Callicarpa kwangtungensis Chun (CK) using high-performance liquid chromatography-electrospray ionization-ion trap-time-of-flight-tandem mass spectrometry (HPLC-ESI-IT-TOF-MS/MS) and a high-performance liquid chromatography-diode array detector (HPLC-DAD). A total of 44 components were characterized from CK, including 13 phenylethanoid glycosides and 17 flavanoids. Furthermore, 14 compounds were identified from CK for the first time. Under the optimal HPLC-DAD chromatographic conditions, six index compounds were quantified in 11 batches of CK on a Phenomenex Luna® C18 column (250 × 4.60 mm; 5 μm) using gradient elution with a solvent system consisting of acetonitrile and 0.1% acetic acid in water at a variable flow rate and detection at 254 nm. All of the calibration curves exhibited good linear relationships (r2 ≥ 0.9999). The RSD values for the intraday precision were less than 1.50% with accuracies between 96.05% and 101.47%. The recoveries of the six analytes were more than 95.01% with RSDs of less than 1.60%. This study presents the first simultaneous characterization and quantitative determination of multiple compounds in CK from locally grown cultivars in China using HPLC-ESI-IT-TOF-MS/MS and HPLC-DAD, which can be employed to standardize the quality of CK.
