Advances in the detection of RNA modification
Abstract
RNA modifications represent a ubiquitous form of chemical labeling that occurs across all RNA species. Current research has identified more than 170 distinct RNA modifications. Understanding the physiological functions and regulatory mechanisms of these modifications is fundamentally dependent on the advanced detection technologies. In this review, we focus on several RNA modifications that have gathered significant research attention in recent years. We present both high-throughput sequencing technologies for transcriptome-wide mapping of these modifications and locus-specific detection methods for characterizing individual sites of interest. The advantages and drawbacks of these methods were also discussed. Finally, we also provide a forward-looking perspective on the evolving landscape of RNA modification research. We aim to provide a comprehensive methodological framework for RNA modification analysis, which may inspire the development of novel detection strategies and facilitate new discoveries in the field of RNA epigenetics.
